What happens when you run:
smbpasswd root
What is the output of:
strings smbd | grep "/smbpasswd"
assuming you are in the directory that contains your smbd binary.
What are the permissions on your smbpasswd file?
Have you verified that you do not have mulitple instances of smbpasswd on
your system?
