Yes, it works.
In the DomU, configure eth0 like you would normally do:
ip addr add 192.168.0.1/32 peer 10.0.0.1 dev eth0
ip route add default via 10.0.0.1
You can then either configure the other domain appropriately or have the P2P
connection terminated in the Dom0 by the associated vif.
I believe that in this case, you need to assign a MAC to the vif and enable
ARP on it. Anyhow, should should remove it from any bridge (xenbr0 by
default).
