Update on this topic. I have just bought a new computer (My previous one was 8 years old and showing its age) and I tried timidity instead of fluidsynth. Midi playback works fine on both the midi player and notation processor in Wine with Pulse Audio enabled.

Why not pulseaudio?
Pulseaudio works great in wine using the modified wine from the Neill Aldur ppa. Tried everything else (padsp, pasuspender, blah) and nothing would work reliably. Some crashes when another application outputs sound, sometimes the sound does not work at all, sometimes it is awfully slow or with latency varying while recording, leading to unpredictable offsets in recorded files. No MIDI devices in wine
I'd want to run the windows editor for my guitar preamp (VOX Tonelab SE) which is connected to the MIDI in and out sockets of my soundcard. The program installs and starts without any problems under wine but it can't find any MIDI ports. MIDI works fine otherwise in Linux - I can read from and write to the sockets with arecordmidi and aplaymidi, but there are no MIDI devices visible

Possible recent bug disables sound in wine - Ubuntu Studio
I have sound problems in Ubuntu Studio, starting yesterday. These may be caused by a Ubuntu bug, but I wanted to run them by wine users and see if anyone else is having a similar problem. Wine why no audio?
Hi. Until Fedora 15, I used qsynth+Fluidsynth+alsa+biab and everything worked fine. But not anymore with Fedora 16. I can't see any midi drivers in band-in-a-box. I'm sure about my setup. Noteworthy Composer crashes when print preview is called.
I have just installed Noteworthy Composer v2.0 It seems to work fine except when I try to call the print preview. The print preview shows up briefly and then Noteworthy shuts itself down. This is almost my first attempt to install anything using Wine. Not quite my first as I installed the Noteworthy Viewer as a trial and that worked once I had sorted out how to get MIDI working. as i do like the pulseaudio server, i do not want to remove it completely. instead, each time i want to run a program thru wine i kill the pulseaudio. can be done by a hotkey or shortcut. ' killall pulseaudio done. Sound works. and when u want pulseaudio to run again, use this ' pulseaudio even simpler. So if you make shortcuts or hotkeys, this is a piece of cake. stop pulseaudio:
